If front and rear tires size of AWD(four wheels drive) is different, and I know that a problem with the drive system.

If I do this change
20x8' front, 245 45ZR 20'
20x10' rear, 275 40ZR 20'

Different width front and rear of the tire, can cause problems with the drive system?

No.  Only need to make sure total height of wheel + tire is within 3% of OEM height.  Also, cannot rotate tires, so plan & budget accordingly.

If tuned:  Take note of the strategy code as you return to stock (including 3 bar MAP to 2 bar MAP) -> take car in & get it serviced -> check strategy code when you get car back -> have tuner update your tune if the strategy code has changed -> reload tune -> ENJOY!

Is it 3% of OEM or 3% of each other? I know one guy on here has 22's on his MKS, be it with smaller tires so maybe its still within 3% of OEM height?

You might want to message bpd1151 as he has run asymmetric front and rear tires/wheels for years without problems.

Both, if the tune is not changed for these parameters.  If the tune is changed to fit your wheel/tire size of choice, and in this case with the staggered setup, the height is of the rears is within 3% (+/-) of the fronts, should be OK as well.

The only negative thing is that if the car's suspension wasn't setup for staggered sizes you will have more understeer when driving the car hard.

I have not had any problems on my flex as I am running 20x8.5 and 20x10 with 275/40 and 315/35.  As stated earlier, rotating is not an option.  But being a badass is.  HAHAHA.   ;-)
